Aerobic and Anaerobic Microbial Degradation of Poly-β-Hydroxybutyrate Produced by Azotobacter chroococcum

Food industry wastewater served as a carbon source for the synthesis of
poly-β-hydroxybutyrate (PHB) by Azotobacter chroococcum . The content of
polymer in bacterial cells grown on the raw materials reached 75%. PHB
films were degraded under aerobic, microaerobic, and anaerobic conditions
in the presence and absence of nitrate by microbial populations of soil, sludges
from anaerobic and nitrifying/denitrifying reactors, and sediment from
a sludge deposit site. Changes in molecular mass, crystallinity, and mechanical
properties of PHB were studied. Anaerobic degradation was accompanied
by acetate formation, which was the main intermediate utilized by
denitrifying bacteria or methanogenic archaea. On a decrease in temperature
from 20 to 5°C in the presence of nitrate, the rate of PHB degradation was
7.3 times lower. Under anaerobic conditions and in the absence of nitrate, no
PHB degradation was observed, even at 11°C. The enrichment cultures of
denitrifying bacteria obtained from soil and anaerobic sludge degraded PHB
films for a short time (3–7 d). The dominant species in the enrichment culture
from soil were Pseudomonas fluorescens and Pseudomonas stutzeri. The rate of
PHB degradation by the enrichment cultures depended on the polymer molecular weight, which reduced with time during biodegradation.

Проведено исследование надмолекулярной структуры волокон полигидроксибутирата, полученных методом гель-формования. Для изученных волокон характерна высокая степень восстанавливаемости
размеров после деформации при значительной степени кристалличности, что типично для так называемых хард-эластиков.
При упругом растяжении образца наряду с рефлексами ромбической ячейки полигидроксибутирата обнаружено появление дополнительного
рефлекса на экваторе рентгенограммы, носящее обратимый характер.

The samples of poly-β-hydroxybutyrate (PHB) with molecular weight 633 kDa were tested for fungicidity and resistance to fungi by estimating the growth rate of test fungi from the genera Aspergillus, Aureobasidium, Chaetomium, Paecilomyces, Penicillium, Trichoderma under optimal growth conditions. PHB film did not exhibit neither fungicide properties, nor the resistance to fungal damage, and served as a good substrate for fungal growth. Changes in molecular weight and mechanical properties of PHB film during biodegradation were studied.
Перспективность использования поли-β-гидроксибутирата (ПГБ) в народном хозяйстве обусловлена его нестойкостью в окружающей среде и возможностью разложения до углекислого газа и воды. Среди факторов, способствующих его разрушению в различных природных средах (гидролиз, механическое или термическое разложение, окисление, фотодеструкция), биодеградация под воздействием бактерий и грибов занимает центральное место. Грибы характеризуются гетеротрофным типом питания, их потребности в питательных субстратах чрезвычайно разнообразны и определяются ферментными системами, которыми они обладают. Многие виды грибов способны усваивать труднодоступные соединения, и это даёт им определённые экологические преимущества в естественной среде обитания. Так как грибы составляют значительную часть микробиоты почвы (Мирчик, 1976), нельзя недооценивать их роль в биодеградации биополимеров. Среди выделенных из почвы микроорганизмов, участвующих в биодеградации полимера полигидроксибутирата, примерно 1/3 составляют грибы
(Maergaert et al., 1992; Matavulj, Molitoris, 1992; Oda et al., 1995).
Способность грибов разрушать ПГБ в природной окружающей среде показана в целом ряде работ. Как правило, грибы-диструкторы выявляли путём высева смывов с образцов биопластика на питательный агар (Matavulj, Molitoris, 1992; Lopez-Liorca et al., 1993; Savencova et al., 2000; Бонарцева и др., 2002). О способности отдельных видов грибов разрушать ПГБ судили по интенсивности их развития и зонам просветления при росте колоний на агазированной среде с полимером (Oda et al., 1995; Козловский и др., 1999). Наиболее часто в литературе как деструкторы ПГБ упоминаются грибы следующих родов: Penicillum, Aspergillus, Paecilomyces, Acremonium, Verticillium, Cephalosporium, Trichoderma.
Целью данной работы было впервые лабораторно испытать полимерную плёнку из ПГБ, полученную нами из клеток Azotobacter chroococcum в Институте биохимии им. А. Н. Баха РАН, на грибостойкость и фунгицидность путём заражения её чистыми культурами плесневых грибов (роды Aspergillus, Aureobasidium, Chaetomium, Paecilomyces, Penicillum, Trichoderma) в оптимальных для их развития условиях; проследить за изменениями молекулярной массы и механических свойств плёнки в процессе биоразрушения.

The content of polymer in bacterial cells grown on the raw materials reached 75%.The films of poly-β-hydroxybutyrate (PHB) were degraded under anaerobic conditions in the presence and absence of nitrate and at low temperatures by microbial populations of sludges from anaerobic and nitrifying/denitrifying reactors, and of sediment from a sludge check. In the presence of nitrate, PHB was degraded to CO2 with formation VFA (acetate, propionate and butyrate) as intermediate products. Then VFA were consumed by denitrifying microorganisms and nitrate was reduced to N2O. Under anaerobic conditions in the absence of nitrate, VFA were formed from PHB rapidly. High concentrations of acetate (to 90 mM) accumulated in medium and suppressed acetoclastic methanogenesis. The rate of PHB degradation at 20° C on all investigated sludges were similar in both the presence and absence of nitrate. PHB degradation was temperature-dependent. In the presence of nitrate, the average rate of PHB degradation and the maximal rate of denitrification decreased 7.3 and 7.1 times, respectively, with a temperature decrease from 20° C to 5°. Under anaerobic conditions without nitrate PHB was not degraded even at 11° C. PHB-degrading microorganisms belong to different morphological types.

The conditions for a maximal poly-β-oxybutyrate synthesis in a poorly active strain of Rhizobium phaseoli have been optimized by the method of mathematical planning of the experiment. Oxygen, sucrose, and potassium nitrate were used as variable factors. As the result of the experiment, the ratios of the factors investigated have been selected so that poly-β-oxybutyrate content in the strain A1 of R. phaseoli reached 80% of dry biomass.

Методом математического планирования эксперимента оптимизированы условия для максимального синтеза поли-β-оксибутирата малоактивным
штаммом Rhizobium phaseoli. В качестве варьируемых факторов взяты кислород, сахароза и нитрат калия. В результате эксперимента
концентрационные соотношения исследуемых факторов были подобраны таким образом, что содержание поли-β-оксибутирата у R. phaseoli, штамм А1
достигало 80% веса сухих клеток.
